Serine323/Serine324 (S323/S324) is major phosphorylation site of the GPR139 Receptor (GPR139). The pS323/pS324-GPR139 antibody detects phosphorylation in response to agonists. S323/S324 phosphorylation is likely to be involved in efficient ligand sequestration.

Upper panel, HEK293 cells stably expressing the GPR139 Receptor (GPR139) and transiently GRK2 (Ratio GPR139 receptor:GRK2 = 10:1) were preincubated with serum-free media. After 2 hours cells were either not exposed or exposed to 100 nM JNJ-63533054 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS323/pS324-GPR139 antibody (7TM0130B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-GPR139 antibody to confirm equal loading of the gel.
