Threonine365/Serine371/Threonine377 (T365/S371/T377) is a major phosphorylation site of the human GPR88 receptor. The pT365/pS371/pT377-GPR88 antibody detects phosphorylation in response to high-efficacy agonists. T365/S371/T377 phosphorylation is a key regulator of human GPR88 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine365/Serine371/Threonine377 phosphorylation of GPR88-Receptor. Upper panel, HEK293 cells stably expressing the GPR88 receptor (GPR88) and transiently GRK2 (Ratio GPR88 receptor:GRK2 = 10:1) were preincubated with serum-free media. After 2 hours cells were either not exposed or exposed to 100 nM of 2-PCCA for 30 minutes. Cells were lysed and immunoblotted with the anti-pT365/pS371/pT377-GPR88 antibody (7TM0123B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-GPR88 antibody (7TM0123N) to confirm equal loading of the gel.
